Rat Lung Fibroblasts (RLF) are derived from the lung of adult Sprague Dawley rat. The cells are cryopreserved at second passage. RLF can be cultured and propagated 10 population doublings. It was found that TGF-beta and PDGF produced by stimulated macrophages are involved in the regulation of the expression of alpha1(1) collagen, of prolyl-4-hydroxylase and lysyloxidase in rat lung fibroblasts1. Antisense calcyclin (S100A6) RNA expression was found to inhibit serum and mechanical strain induced fibroblasts proleferation2. Activation of fibroblasts by TGF-beta(1) by interferon-gamma may have potential therapeutic benefits in bleomycin-induced pulmonary fibrosis3. Furthermore, sulfated proteoglycans play a key role in modulating pulmonary fibroblast extracellular matrix composition and are important mediators of elastolytic injury4, and addition of heparin-binding EGF-like growth factor to pulmonary fibroblasts increases FGF-2 mRNA and protein levels.
