Human Umbilical Vein Smooth Muscle Cells (HUVSMC)
Human umbilical vein smooth muscle cells (HUVSMC) are derived from the umbilical vein. They are cryopreserved at second passage. HUVSMC can be cultured and propagated 6 population doublings. Human umbilical vein actively regulates its vascular tone under the control of pO2. Vasodilation endothelial prostanoids contribute to the hypoxic vasodilation that counteracts the vasoconstricting actions of prostanoids released from HUVSMC. It was found that vasoconstricting prostanoids are released from the endothelium of the human umbilical vein at hyperoxia¹. During preeclampsia a total loss of neuronal nitric oxide synthase (nNOS) protein expression and a significant decrease in mRNA expression were seen. It was found that such loss might alter the regulation of blood flow in the fetal and maternal placental vasculature². Both ouabain and marinobufagenin activate proliferation of the HUVSMC. The concentration ranges of both compounds were found to be below that necessary to induce cytoplasmic ion alterations by sodium pump inhibition³. Mildenberger E, et al. J. Perinat.
