Human
intrathoratic artery smooth muscle cells (HITASMC)
are derived from the intrathoratic artery. They are
cryopreserved at second passage. HITASMC can be
cultured and propagated 8 population doublings.
Proliferation, migration and invasion of SMC are
essential pathogenic processes in the development of
a broad spectrum of cardiovascular disorders. A
Boyden chamber was employed to characterize the
diverging migratory and invasion potential of
proliferating and nonproliferating human artery SMCs.
The differential gene expression of both phenotypes
was analyzed by a cDNA array system. Several
candidate genes were identified1. Left
internal mammary arteries synthesize
endothelium-derived hyperpolarizing factor (EDHF), a
short-lived K(+) channel activator that persists
after inhibition of nitric oxide (NO) and
prostaglandin synthesis. The identity of EDHF is
identified as 11,12-epoxyeicosatrienoic acid
(11,12-EET) produced by an EDHF synthase CYP450-2C,
and accounting for about 40% net endothelial cell
relaxation2. Expression of non-degradable
IkappaB-alpha might have therapeutic potential in
both vascular inflammatory reaction and SMC
proliferation3.Using HITSMC, it was found
that amolodipine can alter Ca2+ mobilization and
inhibit voltage-dependent Ca2+ influx and the
resulting ERK 1/2 activation4. In human
vascular smooth muscle cells of both saphenous vein
and internal thoracic artery, UTP was found to act
as an anti-proliferative regulator5.
